Product Specific Information

The HA58 monoclonal antibody reacts with human CD54 (ICAM-1), a 90–110 kDa transmembrane glycoprotein expressed by monocytes, lymphocytes, and endothelial cells. Expression is upregulated upon lymphocyte activation, and interaction with CD11a is critical in inflammatory responses.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Normal human peripheral blood cells, ≤5 µL (0.6 µg)/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test (optimize empirically).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL of cell sample for single-color controls.
• NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures minimizing spillover and spread.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein with five immunoglobulin-like extracellular domains, a transmembrane region, and a cytoplasmic domain. It has seven potential N-linked glycosylation sites and shares homology with ICAM-2 and ICAM-3.

• Rhinovirus receptor

Expression: Activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, B/T lymphocytes (low), monocytes, macrophages, dendritic cells, neutrophils, and some carcinoma/melanoma cells.
Soluble ICAM-1: Detectable in plasma; elevated in inflammatory syndromes.
