Product Specific Information

The N2B2 monoclonal antibody reacts with mouse TNF-related apoptosis-inducing ligand (TRAIL), a member of the TNF superfamily. TRAIL is not detected on the surface of freshly isolated T, B, or NK cells, but can be induced preferentially on CD3⁻ NK1.1⁺ NK cells after stimulation with IL-2 or IL-15.
N2B2 inhibits IL-2- or IL-15-activated NK cell cytotoxicity against mouse fibrosarcoma L929 target cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse CD253 transfected cells; usable at ≤0.125 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best results.

Target Information

TRAIL is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells while sparing normal cells. TRAIL binds to several TNF receptor superfamily members, including:

  • TNFRSF10A/TRAILR1
  • TNFRSF10B/TRAILR2
  • TNFRSF10C/TRAILR3
  • TNFRSF10D/TRAILR4
  • TNFRSF11B/OPG

Its activity may be modulated by decoy receptors (TRAILR3, TRAILR4, OPG) that cannot induce apoptosis. Binding of TRAIL to its receptors triggers MAPK8/JNK, caspase 8, and caspase 3 activation.
Receptors include death receptors DR4 and DR5, and decoy receptors DcR1 and DcR2. TRAIL is a type II membrane protein expressed in various tissues, inducing apoptosis and NF-κB activation.
