Oxygen radicals damage chromosomal DNA, leading to cell death and mutations. While much attention has focused on repairing 8-oxo-G lesions, many other DNA sites are damaged by free radicals. The human NTH1 repair protein acts on a wide range of these oxidatively damaged DNA sites. As a homologue of E. coli Endonuclease III, NTH is a DNA glycosylase with apurinic/apyrimidinic lyase activity. It exhibits broad substrate specificity, targeting various ring saturation and fragmentation products of pyrimidines. Research indicates that NTH plays a crucial role in removing oxidative base lesions in mitochondrial DNA.
