Product Specific Information

For reconstitution, we recommend adding 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments using this carrier-free antibody, perform an additional desalting step (Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The transcription of many RNA polymerase II-dependent genes requires Spt3, a member of the S. cerevisiae SAGA complex. Transcription from delta sequences, the long terminal repeats that flank yeast Ty elements, requires the yeast SPT3 gene.
Spt3 and Spt20 cooperate to recruit TATA-box binding protein (TBP) to the core promoter, enabling TBP binding to SAGA-dependent promoters. Null mutations in the Spt3 gene cause defects in sporulation, diploid filamentous growth, and haploid invasive growth, highlighting its role in mating and developmental pathways.
At some gene promoters, including yeast HO, HIS3, and TRP3, Spt3 inhibits TBP binding, reducing transcription. This repression can be counteracted by GCN5, another SAGA complex member, which promotes TBP/TFIIA complex formation via histone acetylation.
