Target Information
Follistatin-like protein 1 (FSTL1) is a widely-expressed extracellular glycoprotein belonging to the osteonectin (BM-40/SPARC) family. It contains both a follistatin-like and extracellular calcium-binding domain. Initially identified as a TGF-beta-inducible protein in mouse osteoblasts, FSTL1 is involved in various cellular functions including proliferation, differentiation, apoptosis, and migration. It also plays roles in embryonic development, inflammation, angiogenesis, tumorigenesis, and immune disease pathogenesis.
FSTL1 expression is highly conserved across mammals and can be upregulated by innate immune mediators such as TLR4 agonists and IL-1β via NFκB signaling, promoting secretion of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, IL-8). While mesenchymal cells produce FSTL1, hematopoietic cells typically do not. At inflammation sites, macrophages and monocytes can uptake FSTL1, leading to caspase-1 activation and IL-1β processing. Overexpression of FSTL1 contributes to rheumatoid arthritis and osteoarthritis progression, whereas decreased serum levels are linked to ovarian and endometrial cancer development.
