Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
Suggested positive controls include skin, tonsil, or squamous epithelium.

Target Information

N-cadherin is a 140 kDa transmembrane protein belonging to a family of calcium-dependent cell adhesion molecules. These proteins mediate intercellular adhesion and regulate morphogenetic movements during development.
N-cadherin function depends on its interaction with the actin cytoskeleton through catenin proteins. The stability of this complex is regulated by phosphorylation and dephosphorylation of beta-catenin.
N-cadherin contains five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail. It plays a key role during gastrulation and is essential for establishing left-right asymmetry.
