Target Information
RNF11 is an essential component of a ubiquitin-editing protein complex that includes TNFAIP3, ITCH, and TAX1BP1. This complex regulates inflammatory signaling pathways by promoting the association of TNFAIP3 with RIPK1 following TNF stimulation. TNFAIP3 deubiquitinates Lys-63 polyubiquitin chains on RIPK1 and catalyzes the formation of Lys-48 polyubiquitin chains, leading to RIPK1 proteasomal degradation and termination of NF-kappa-B activation. RNF11 also recruits STAMBP to the E3 ubiquitin-ligase SMURF2 for ubiquitination, resulting in its degradation by the 26S proteasome.
