Target Information
Proposed scaffold protein implicated in diverse physiological processes, partly linked to regulation of ubiquitination of specific cellular proteins. Modulates activity of cullin-RING E3 ubiquitin ligase (CRL) complexes by displacing CAND1; promotes CRL E3 activity and dissociates CAND1 from CUL1 and CUL2. Promotes ubiquitination and proteasomal degradation of NF-kappa-B subunit RELA, down-regulating NF-kappa-B activity. Regulates membrane expression and ubiquitination of SLC12A2, modulating Na(+) transport in epithelial cells. Promotes localization of SCNN1D to recycling endosomes and CFTR cell surface expression through regulation of its ubiquitination. Down-regulates SOD1 activity by interfering with homodimerization. Plays a role in copper ion homeostasis and ATP7A trafficking between trans-Golgi network and peripheral vesicles, dependent on CCC and WASH complex cooperation. Binds one copper ion per monomer and phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2). Involved in HIF1A-mediated transcription regulation by competing with ARNT/Hif-1-beta for HIF1A binding, reducing DNA binding and transcriptional activation.
