Target Information

Proposed scaffold protein implicated in diverse physiological processes, partly linked to its ability to regulate ubiquitination of specific cellular proteins. Modulates activity of cullin-RING E3 ubiquitin ligase (CRL) complexes by displacing CAND1, promoting CRL E3 activity and dissociating CAND1 from CUL1 and CUL2. Promotes ubiquitination and proteasomal degradation of NF-kappa-B subunit RELA, thereby down-regulating NF-kappa-B activity. Regulates membrane expression and ubiquitination of SLC12A2 and modulates Na(+) transport in epithelial cells by controlling ENaC subunit expression and ubiquitination via NEDD4L. Promotes localization of SCNN1D to recycling endosomes and CFTR cell surface expression through regulation of its ubiquitination. Down-regulates SOD1 activity by interfering with homodimerization. Plays a role in copper ion homeostasis and ATP7A trafficking between the trans-Golgi network and peripheral vesicles, depending on association within the CCC complex and cooperation with the WASH complex on early endosomes. Binds one copper ion per monomer and may facilitate biliary copper excretion within hepatocytes. Also binds phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) and regulates HIF1A-mediated transcription by competing with ARNT/Hif-1-beta for HIF1A binding, reducing DNA binding and transcriptional activation by HIF-1.
