Target Information

Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing. This process can affect gene expression and function in multiple ways, including:

  • Altering mRNA translation by changing codons and amino acid sequences
  • Modifying pre-mRNA splicing through splice site recognition changes
  • Affecting RNA stability via nuclease recognition sequence modification
  • Influencing genetic stability in RNA virus genomes
  • Regulating RNA structure-dependent activities such as microRNA production and protein-RNA interactions

ADAR can edit both viral and cellular RNAs, either at multiple (hyper-editing) or specific sites. Cellular RNA substrates include BLCAP, GRIA2, HTR2C, and GABRA3, where site-specific RNA editing alters protein function.

Viral RNA substrates include HCV, VSV, MV, HDV, and HIV-1. ADAR1 exhibits either proviral or antiviral effects that may be editing-dependent or independent. For example:

  • Inhibits HCV replication via RNA editing
  • Enhances replication of MV, VSV, and HIV-1 via suppression of EIF2AK2/PKR activation
  • Stimulates HIV-1 infectivity through editing in the 5′UTR and coding regions
  • Enhances HDV replication by A-to-I editing at amber/W site, promoting large delta antigen synthesis

However, excessive ADAR1 expression can inhibit HDV replication.
