Target Information
The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some members are active cysteine protease inhibitors, while others have lost or never acquired this activity.
There are three inhibitory families within the superfamily:
- Type 1 cystatins (stefins)
- Type 2 cystatins
- Kininogens
Type 2 cystatin proteins are cysteine proteinase inhibitors found in various human fluids and secretions.
The cystatin locus on chromosome 20 contains most type 2 cystatin genes and pseudogenes. This gene encodes a type 2 salivary cysteine peptidase inhibitor (S-type cystatin), highly expressed in saliva, tears, and seminal plasma.
Its role is likely protective, with demonstrated antibacterial and antiviral activity.
