Target Information

SRC2 (steroid receptor coactivator 2), also known as nuclear receptor coactivator 2 (NCOA2) or TIF2, aids in the function of nuclear hormone receptors. These receptors act as conditional transcription factors regulating gene expression involved in cell growth, development, and homeostasis. Members of this receptor superfamily are structurally characterized by three domains:

  • N-terminal transcriptional activation domain
  • Central DNA-binding domain
  • C-terminal hormone-binding domain

Before hormone binding, steroid receptors remain inactive in a complex with heat-shock proteins. Hormone binding induces conformational changes, causing dissociation from inhibitory complexes and enabling receptor dimerization, DNA binding, and transcriptional activation. Coactivator proteins such as SRC2 are required for efficient transcription initiation.
