Target Information
The CDKN2A gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants have been identified, two encoding isoforms that inhibit CDK4 kinase. Another transcript encodes an alternate open reading frame (ARF) that produces a structurally distinct protein functioning as a stabilizer of tumor suppressor protein p53 by interacting with and sequestering MDM2, which mediates p53 degradation. Despite structural differences, both CDK inhibitor isoforms and the ARF product contribute to cell cycle G1 control through regulation of CDK4 and p53. CDKN2A is frequently mutated or deleted in various tumors and functions as a critical tumor suppressor gene.
