Target Information
KLRB1 (Killer cell lectin like receptor subfamily B, member 1, CD161) protein is a type II membrane protein with an external C terminus. It is expressed on nearly all NK cells and a subset of CD3⁺ T cells.
KLRB1 forms a homodimeric cell surface structure (40–44 kDa) and plays an inhibitory role in NK cell activity. Activation triggers acid sphingomyelinase/SMPD1 and increases intracellular ceramide, along with AKT1/PKB and RPS6KA1/RSK1 kinase activation, promoting T cell proliferation by anti-CD3 stimulation.
It binds to Gal-α(1,3)-Gal and N-acetyllactosamine epitopes, as well as CLEC2D/LLT1 ligand, inhibiting NK cell-mediated cytotoxicity and interferon-γ secretion.
The protein includes extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
