Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 125 kDa transmembrane protein expressed by mature macrophages. F4/80 is a key marker for macrophage populations and is also found in Langerhans and Kupffer cells. Expression begins in early myeloid development and is upregulated by M-CSF stimulation. Certain cytokines can downregulate F4/80 expression, leading to subpopulations lacking the antigen in lymphoid environments.

Applications Reported:
BM8 has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and bone marrow cells. Recommended ≤ 0.5 µg per test (100 µL final volume). Cell counts may range from 10⁵ to 10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein of the EGF TM7 family, sharing 68% amino acid identity with human EMR1. Expression is heterogeneous and varies with macrophage maturation and activation. Found on tissue macrophages such as Kupffer cells, Langerhans, microglia, and macrophages in spleen, thymus, and bone marrow. Also detected on some dendritic cells but absent in T cell zones of spleen and lymph nodes. F4/80 may play a role in generating efferent CD8⁺ regulatory T cells.
