Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase that plays an essential role in the T-cell activation pathway.
It consists of two subunits:

  • Calcineurin A (CnA): 60 kDa catalytic subunit, homologous to protein phosphatases 1 and 2A
  • Calcineurin B (CnB): 19 kDa regulatory subunit with four EF-hand calcium-binding sites

Calcineurin-mediated dephosphorylation of NF-AT is essential for its activation, nuclear translocation, and early gene expression in T-cells.
Immunosuppressive drugs such as cyclosporin A and FK506 inhibit NF-AT activation by forming complexes that bind to both CnA and CnB, blocking substrate access to CnA’s catalytic domain.
