Product Specific Information

The 34-1-2S monoclonal antibody reacts with mouse MHC class I molecules H-2Kd and H-2Dd. It also cross-reacts with Kb, s, r, q, and p haplotypes.
This antibody is validated for flow cytometric analysis of mouse splenocytes and can be used at ≤0.4 µg per test (in 100 µL final volume). Cell concentration may vary from 10⁵–10⁸ cells/test and should be optimized empirically.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains for viability analysis with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence performance and reduced spectral spillover.

Target Information

MHC Class I molecules are involved in antigen presentation to CD3/TCR and CD8-expressing T cells.
These antibodies are valuable tools in Immunopeptidomics, aiding in neoantigen identification and characterization via HPLC-tandem MS analysis.
