Target Information

Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing. This process can affect gene expression and function through:

  • mRNA translation (codon changes leading to altered proteins)
  • pre-mRNA splicing (splice site recognition changes)
  • RNA stability (nuclease recognition sequence changes)
  • genetic stability in RNA virus genomes
  • RNA structure-dependent activities (microRNA production, protein-RNA interactions)

ADAR edits both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing).
Cellular RNA substrates: BLCAP, GRIA2, HTR2C, GABRA3
Viral RNA substrates: HCV, VSV, MV, HDV, HIV-1

ADAR exhibits variable effects depending on the virus type:

  • Antiviral: HCV (editing-dependent)
  • Proviral: HDV, MV, VSV, HIV-1 (editing-dependent or independent)

It modulates viral replication through mechanisms involving EIF2AK2/PKR suppression and A-to-I editing at specific viral RNA sites.
