Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing. This process can affect gene expression and function through:
- mRNA translation (codon changes leading to altered proteins)
- pre-mRNA splicing (splice site recognition changes)
- RNA stability (nuclease recognition sequence changes)
- genetic stability in RNA virus genomes
- RNA structure-dependent activities (microRNA production, protein-RNA interactions)
ADAR edits both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing).

Viral RNA substrates: HCV, VSV, MV, HDV, HIV-1
ADAR exhibits variable effects depending on the virus type:
