Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha)는 수용체형 막단백질로, 과립구, 수지상세포(DCs), 대식세포, 비만세포, 조혈줄기세포 등 골수 계통 세포에서 발현됩니다.
이 단백질은 EGFR, PDGFR, src, 인슐린 수용체 등 여러 활성화된 티로신 키나아제의 기질로 작용하며, 수용체 티로신 키나아제 신호경로의 음성 조절에 관여합니다.
SIRP alpha는 CD47과 결합하여 세포 내 ITIM 영역의 티로신 인산화를 유도하고, SHP-1 및 SHP-2 인산화효소의 활성화를 매개합니다.
이 상호작용은 대식세포의 식작용 억제, T세포 및 DC의 양방향 활성화 등 다양한 면역 조절 과정에 관여합니다.
암세포는 CD47을 과발현하여 SIRP alpha를 활성화하고, 대식세포에 의한 암세포 제거를 억제하는 것으로 알려져 있습니다.
