Target Information
Epidermal Growth Factor (EGF) binds to the EGF receptor (EGFR), a 170 kDa protein kinase. Activation of EGFR initiates diverse cellular pathways in response to environmental stimuli. Upon EGF binding, EGFR forms homo- or heterodimers with other family members, each activating distinct signaling cascades via SH2-containing effector proteins.
EGF is widely expressed in kidney, brain, prostate, and salivary glands, acting as a potent mitogenic factor. The phosphorylated receptor recruits adapter proteins like GRB2, triggering downstream signaling cascades such as RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, and STAT pathways.
EGF may also activate NF-κB signaling. Defects in the EGF gene are linked to hypomagnesemia type 4, and dysregulation has been associated with cancer growth and progression.
