Target Information
IA-2 (insulinoma-associated protein 2, ICA512, PTPRN) is a member of the transmembrane protein tyrosine phosphatase (PTP) family located in secretory granules of neuroendocrine cells. The IA-2 protein (979 amino acids) includes luminal, transmembrane, and cytoplasmic domains. Despite being a PTP family member, IA-2 lacks phosphatase activity due to substitutions at critical catalytic residues. IA-2 and IA-2β are major autoantigens in type 1 diabetes mellitus. Upon granule exocytosis, the IA-2 cytoplasmic domain translocates to the nucleus, enhancing phosphorylated STAT5/STAT3 levels, which promotes insulin gene transcription and granule biogenesis. IA-2 signaling also regulates pancreatic β-cell proliferation through STAT-mediated cyclin D control.
