Target Information
Protein C is a vitamin K-dependent plasma glycoprotein that is cleaved to its activated form by the thrombin-thrombomodulin complex. Activated Protein C contains a serine protease domain and functions in degradation of the activated forms of coagulation factors V and VIII. It is produced in the liver and composed of two polypeptide chains. The 62 kDa proenzyme is activated by thrombin bound to an endothelial surface receptor, forming a 55 kDa active enzyme that cleaves factor Va and VIIIa to inhibit blood coagulation. Normal human plasma concentration of active Protein C is approximately 1–3 ng/mL, while the proenzyme concentration is around 3 µg/mL. Mutations in the Protein C gene are associated with thrombophilia, neonatal purpura fulminans, and recurrent venous thrombosis.
