Description:
The R12-31 monoclonal antibody reacts with mouse RANK (Receptor Activator of NF-kappaB), a member of the TNFR superfamily. RANK ligand (RANKL/TRANCE/OPGL) binds to RANK on dendritic cells and upregulates anti-apoptotic protein BcL-XL, indicating a role in dendritic cell survival. The cytoplasmic domain of RANK interacts with TRAF2, TRAF5, and TRAF6. Overexpression of RANK activates NF-kappaB and JNK pathways. RANK–TRAF6 interaction activates NF-kappaB, while JNK activation is mediated via TRAF2 binding.

Applications Reported:
The R12-31 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse RANK-transfected cells. Use ≤1 µg per test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number can range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal assay performance.

Target Information

RANK is a member of the tumor necrosis factor receptor (TNFR) family. The full-length mouse RANK cDNA encodes a type I transmembrane protein with extracellular and cytoplasmic domains. The extracellular domain contains two potential N-linked glycosylation sites and four cysteine-rich repeats, characteristic of the TNFR family. Human and murine RANK share 81% amino acid identity in their extracellular domains.
