Product Specific Information

The monoclonal antibody 17-206 recognizes the human blood group antigen H (O) type 1, primarily found in epithelial and secretory cells. The precursor H antigen is modified by saccharide addition to carbohydrate chains of proteins and lipids on erythrocytes and epithelial cells. The type and sequence of saccharides determine the blood group subtype (A, B, AB, or O), mediated by glycosyl transferases.

Evidence suggests that blood group antigen H expression is reduced or absent in tumor cells, correlating with disease progression, implying a role in cell proliferation, adhesion, and angiogenesis. The 17-206 antibody does not recognize blood group antigen H (O) type 2.

Applications Reported: Suitable for immunohistochemical staining of frozen and formalin-fixed paraffin-embedded tissue sections, and immunocytochemistry.
Applications Tested: Tested by IHC on formalin-fixed human tissue at ≤10 µg/mL. Titrate for optimal performance.

Target Information

The H antigen is a precursor to ABO blood group antigens, found in individuals of all common blood types. The Bombay phenotype (hh) lacks H antigen and therefore also lacks A or B antigens, producing isoantibodies to H, A, and B. Such individuals can only receive blood from other hh donors.

The H antigen production is controlled by the H locus on chromosome 19, which encodes a fucosyltransferase responsible for H antigen synthesis. The H antigen is a carbohydrate sequence mainly linked to protein, with a minor fraction attached to ceramide.
