Target Information

During mitosis, the cell’s microtubule (MT) network forms the mitotic spindle, enabling segregation of paired chromosomes. Kinetochores (KT) on centromeric chromatin are essential for proper chromosome-spindle attachment.
The spindle and kinetochore complex (SKA), composed of SKA1, SKA2, and SKA3, is crucial for stable KT-MT interactions and timely anaphase onset.
Depletion of SKA1 or SKA2 by siRNA causes loss of both proteins from the KT without affecting overall KT structure. Cells lacking the SKA complex experience prolonged metaphase-like delay, underscoring its role in spindle checkpoint silencing and metaphase plate maintenance.
