Target Information
Protein tyrosine phosphatases (PTPs) are enzymes that remove phosphate groups from phosphorylated tyrosine residues on proteins. Together with tyrosine kinases, they regulate the phosphorylation state of key signaling molecules, including the MAP kinase family.
Dual specificity phosphatases (DSPs) are a subclass of the PTP superfamily that dephosphorylate both phosphothreonine and phosphotyrosine residues. The VH1 gene in vaccinia virus is a prototypical DSP expressed during late-stage infection. The shallow active site of VHR allows hydrolysis of phosphorylated serine, threonine, or tyrosine residues, unlike the deeper active site of typical PTPs which restricts specificity to phosphotyrosine.
