Target Information
FAS (Fas, CD95, APO-1)은 TNFR (tumor necrosis factor receptor) 초가족에 속하는 46 kDa 막단백질로, 세포사멸 수용체로 기능합니다. 또한 약 26 kDa의 가용성 형태로도 존재합니다. FAS 자극은 FADD(FAS-associated death domain) 단백질의 응집, 사멸 유도 신호 복합체(DISC) 형성 및 caspase 활성화를 유도합니다. 이 복합체 내 caspase의 자동 절단은 세포사멸(apoptosis)을 유도하는 하위 신호전달을 촉발합니다. FAS는 NF-kappaB, MAPK3/ERK1, MAPK8/JNK 경로를 활성화하며, 정상 섬유아세포 및 T세포의 증식 신호 전달에도 관여합니다. 최소 8개의 전사체 변이체가 보고되었으며, 막관통 도메인이 결여된 형태는 완전 길이 단백질의 세포사멸 유도를 억제할 수 있습니다. FAS/FAS ligand 시스템은 AIDS, 간염, 암 등 다양한 인간 질환과 관련이 있으며, 항바이러스 면역 반응에서 중요한 역할을 하는 것으로 알려져 있습니다.
