Product Specific Information

The GK1.5 monoclonal antibody recognizes the mouse CD4 molecule, a 55 kDa surface receptor expressed by thymocytes, mature T cell subpopulations, and dendritic cells. CD4 binds MHC class II molecules on antigen-presenting cells and plays a critical role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.2 µg/test (100 µL final volume). Cell concentration: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Improved formulation over CellBlox. Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktail/master mix before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton™ nucleic acid dye structures engineered for controlled fluorescent signatures minimizing spillover and spread.
Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).

Note:
Internal testing shows NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice. No such staining observed in BALB/c or C57BL/6 strains.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. Expressed mainly on T-helper lymphocytes and also on monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 facilitates T-cell activation via its association with the TCR complex and Lck kinase. It may mediate neuronal damage in immune-related CNS diseases.
