Product Specific Information

The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity alone but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (γ chain). It plays a role in lymphocyte differentiation, activation, and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of human peripheral blood cells; use 5 µL (0.1 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails prior to combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2RA, IL2RB, and IL2RG chains form the high-affinity IL-2 receptor complex. Homodimeric IL2RA forms a low-affinity receptor, while IL2RB homodimers form a medium-affinity receptor. IL2 is primarily produced by mature T cells and regulates growth, differentiation, and apoptosis. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency and immune dysregulation.
