Product Specific Information

The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. By itself, CD25 binds IL-2 with low affinity, but associates with CD122 (IL-2Rβ) and CD132 (common γ chain) to form the high-affinity IL-2 receptor. CD25 plays a role in lymphocyte differentiation, activation, and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Use 5 µL (0.1 µg) per test in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL of CellBlox Plus per sample, regardless of panel size.
• For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor component involved in T and B lymphocyte proliferation. The high-affinity IL-2 receptor complex consists of IL2RA, IL2RB, and IL2RG subunits. IL2 plays a critical role as a growth and differentiation factor and regulator of cell death. It promotes B cell proliferation, enhances NK cell activity, and inhibits granulocyte-macrophage colony formation. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency.
