Target Information
BRCA1-associated protein-1 (BAP1)는 BRCA1의 RING finger domain과 상호작용하는 단백질입니다. 인간 BAP1 단백질의 N-말단 240 아미노산은 ubiquitin C-terminal hydrolases (UCHs)와 상동성을 가지며, 이는 ubiquitin의 단백질분해 처리에 관여하는 thiol protease입니다.
BAP1은 산성 영역, 고전하성 C-말단 영역, 2개의 핵내 위치 신호(NLS)를 포함합니다. BAP1과 BRCA1은 세포 내에서 함께 존재하며, 유방암 세포 성장 억제에 관여합니다.
Northern blot 분석 결과, BAP1은 대부분의 인체 조직에서 4-kb mRNA로 발현되며, 고환에서는 4.8-kb 전사체가 특이적으로 발현됩니다. 또한 BAP1과 BRCA1은 생쥐 유방 발달 및 재형성 과정에서 공동 발현됩니다.
BAP1 유전자는 3p21.3에 위치하며, 이 부위는 유방암 및 폐암에서 이형접합성 소실이 자주 관찰되는 영역입니다. 폐암 세포주에서는 BAP1의 동형접합 재배열 및 결실이 발견되며, BAP1은 BRCA1 성장 조절 경로에서 종양 억제 유전자로 기능하는 것으로 추정됩니다.
