Product Specific Information
- Antibody tested in direct ELISA and IHC (PFA fixed).
- For IHC, incubate antibody for 30 min at room temperature.
- For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at RT for 20 min.
Target Information
ZAP-70 is a cytosolic protein tyrosine kinase belonging to the Syk family, expressed exclusively in T-cells and natural killer cells. It is essential for T-cell receptor activation. Upon TCR engagement, ZAP70 is phosphorylated on tyrosines 292, 315, and 319 in the interdomain B region. Phosphorylation at tyrosines 315 (Vav binding site) and 319 (Lck binding site) enhances signal transduction, while phosphorylation at tyrosine 292 attenuates activation.
