Background

Ataxin 1 (ATXN1) is associated with autosomal dominant cerebellar ataxias (ADCA), a group of neurodegenerative disorders characterized by progressive degeneration of the cerebellum, brain stem, and spinal cord. ADCA is divided into three groups (types I–III). Type I includes several spinocerebellar ataxia (SCA) subtypes—SCA1, 2, 3, 4, and 6—linked to different chromosomes. Type II (SCA7) presents with retinal degeneration, while type III (SCA5) is a pure cerebellar syndrome. Many SCA genes contain CAG repeat expansions in their coding regions, leading to elongated polyglutamine tracts in the corresponding proteins. These expanded repeats are unstable and tend to increase in size upon transmission, contributing to disease progression.
