Background
The ret proto-oncogene (RET) encodes a receptor tyrosine kinase, a cell-surface molecule that transduces signals for cell growth and differentiation. It plays a crucial role in neural crest development and can undergo oncogenic activation through cytogenetic rearrangement. Mutations in this gene are associated with multiple endocrine neoplasia type IIA and IIB, Hirschsprung disease, and medullary thyroid carcinoma. Two transcript variants encoding different isoforms have been identified, and additional variants have been described though their biological validity remains unconfirmed.
