Background

This gene is the cellular homolog of the viral raf gene (v-raf). The encoded protein is a MAP kinase kinase kinase (MAP3K), functioning downstream of Ras family GTPases. Activated RAF1 phosphorylates and activates MEK1/2, which in turn activate ERK1/2. These kinases regulate gene expression related to cell cycle, apoptosis, differentiation, and migration.
Mutations in this gene are associated with Noonan syndrome 5 and LEOPARD syndrome 2.
