Target Information
A recently identified protein, termed carboxyl terminus of hsc70-interacting protein (CHIP), interacts with both the constitutive form of hsc70 and the stress-inducible form, hsp70. This 35 kDa cytoplasmic protein is highly expressed in striated muscle and across a broad range of cultured tissues. Through immunoprecipitation experiments, CHIP has been shown to bind directly to the carboxyl terminus of hsc70 and hsp70, decreasing ATPase activity and reducing overall chaperone efficiency. CHIP also functions as an E3 ubiquitin ligase in the ubiquitin-proteasome system, containing a U-box domain and acting in conjunction with hsc70 and hsp90.
