Target Information
EIF2AK2 (PKR)는 eIF2-alpha의 Ser51을 인산화하는 4가지 키나아제 중 하나로, 다양한 환경 스트레스에 반응합니다. PKR은 바이러스 감염 시 생성되는 이중가닥 RNA에 의해 직접 활성화되어 항바이러스 방어에 중요한 역할을 합니다. 또한 IFN 유도 선천 면역 반응, 바이러스 유도 세포자멸사, 세포 성장 및 분화 조절에 관여합니다. PKR은 Fas 발현을 상향 조절하여 FADD/Caspase-8 경로를 통해 세포자멸사를 유도합니다. 바이러스 감염 시 PKR은 세포 생존 및 사멸 경로를 순차적으로 활성화하며, NF-kappaB, p53, p38, PDGF 등의 신호 경로를 조절합니다. PKR은 종양 억제 및 악성화에 관련되어 있으며, 일부 바이러스는 PKR 억제 단백질(NS1 등)을 통해 방어 기전을 회피합니다. 최근에는 알츠하이머, 헌팅턴병, 근위축성 측삭경화증 등 신경퇴행성 질환과의 연관성이 보고되었습니다.
