Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification by immobilized metal affinity chromatography.
Epitope tagging allows detection of recombinant proteins for which no specific antibody is available. His-tagged proteins containing histidine clusters are purified through metal chelate chromatography, particularly with Ni²⁺-NTA adsorbents due to strong and selective chelation affinity.
His tag-specific antibodies are used for detection and co-immunoprecipitation of tagged proteins.
