Product Specific Information
- Immunohistochemistry (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating sections in 10 mM Tris Buffer with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes. - Western Blot: Use 1–2 µg/mL for 2 hours at room temperature.
Target Information
CD81 (TAPA-1, target of anti-proliferative antibody-1) is a member of the tetraspanin family expressed on most nucleated cells, especially germinal center B cells. It forms complexes with tetraspanin proteins, integrins, coreceptors, and MHC molecules, influencing adhesion, morphology, activation, proliferation, and differentiation of B and T cells.
In muscle, CD81 promotes cell fusion and myotube maintenance, and acts as a receptor for the hepatitis C virus. Like other tetraspanins (CD9, CD37, CD53, CD63, CD82), CD81 is characterized by four hydrophobic domains mediating signal transduction events that regulate cell development and motility.
CD81 associates with CD19, CD21, Leu 13, and integrins, and antibodies to CD81 exhibit anti-proliferative effects on various lymphoid cell lines. It is localized in a tumor-suppressor gene region and considered a candidate gene for malignancies.
