Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the HSP70 family. It is an endoplasmic reticulum (ER) resident protein that assists in the folding and assembly of newly synthesized secretory and membrane proteins. GRP78 also interacts with misfolded proteins, marking them for degradation. The conserved KDEL sequence at its C-terminus ensures ER protein retention via the KDEL receptor.
GRP78 plays a key role in protein folding, degradation of misfolded proteins, and maintenance of cellular homeostasis. It is implicated in neuroprotection under oxidative stress and serves as a biomarker in hypoglycemia. Reduced GRP78 levels are observed in Alzheimer’s disease and certain PS1 gene mutations. Induction of GRP78 has been linked to antitumor drug resistance.
