Target Information

The expression pattern of the Prox1 homeobox gene suggests that it plays a role in various embryonic tissues, including the lens. Prox1 mRNA is present in many human tissues, with the highest level observed in the lens. Homozygous Prox1⁻/⁻ mice die at midgestation due to multiple developmental defects, including effects on lens development. Prox1 inactivation causes abnormal cell proliferation, downregulation of Cdkn1b and Cdkn1c, misexpression of E-cadherin, and excessive apoptosis, leading to improper lens cell elongation and hollow lens formation.

Prox1 is expressed in a subset of endothelial cells that form the lymphatic system and acts as a required regulator of lymphatic development. It is also necessary for hepatocyte migration in mice; loss of Prox1 results in smaller livers with reduced hepatocyte clusters. In the embryonic mouse retina, Prox1 regulates progenitor cell cycle exit, influencing cell proliferation and fate determination.
