Target Information

Matrix metalloproteinases (MMPs) are proteolytic enzymes that degrade components of the extracellular matrix. They share a common activation mode, a conserved metal-binding active site sequence, and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs play essential roles in angiogenesis regulation.

MMP2 is synthesized as a 631-amino-acid proenzyme, activated by cleavage of its first 80 amino acids. It includes propeptide, catalytic, and hemopexin domains. The MMP family comprises at least eighteen secreted and membrane-bound zinc endopeptidases, collectively capable of degrading all extracellular matrix components such as collagens, fibronectin, laminin, and basement membrane glycoproteins.

MMP2 is involved in tissue remodeling, and mutations in the MMP2 gene are associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP2 have been identified.
