RNF2 (E3 ubiquitin-protein ligase RING2)는 히스톤 H2A의 Lys-119 단일 유비퀴틴화를 매개하는 리가아제로, 히스톤 코드 및 유전자 조절에 핵심적인 역할을 수행합니다. BMI1/PCGF4에 의해 리가아제 활성이 증가하며, H2AK119Ub는 전사 억제에 관여하고 암컷 포유류의 X 염색체 불활성화 과정에도 참여합니다. 또한 RNF2는 Polycomb group (PcG) 단백질 복합체 PRC1의 필수 구성 요소로, 발달 과정 전반에서 Hox 유전자 등 다양한 유전자의 전사 억제 상태를 유지하는 데 필요합니다. 이 복합체는 히스톤 변형 및 염색질 재구성을 통해 유전자 발현 상태를 안정적으로 유지합니다.
