Product Specific Information

The eBio4B10 monoclonal antibody reacts with mouse and human T-bet, a Th1-specific T-box transcription factor critical for Th1 CD4⁺ T cell lineage development.
T-bet expression is induced by IFN-γ and regulates IFN-γ, IL-12Rβ, and IL-2 expression. It also plays a role in class-switch recombination in B-cells.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated for intracellular staining and flow cytometry using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).
Recommended use ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Titrate for optimal performance.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.

Target Information

T-bet (TBX21) is a T-cell specific transcription factor with a T-box DNA-binding domain. It controls interferon-gamma expression and directs Th1 lineage development from naive precursors.
T-bet cooperates with Stat4 in programming chromatin architecture for Th1 gene expression.
Genetic variations are linked to aspirin-induced asthma and nasal polyps.
