Description:
The 1D6 monoclonal antibody reacts with human BLyS (B lymphocyte stimulator), also known as BAFF (B cell activating factor from the TNF family), TALL-1, THANK, and zTNF4. BLyS is a type II membrane protein existing in both membrane-bound and soluble forms. Expression on monocytes is regulated by IFN-γ. BLyS functions as a potent B cell growth factor and costimulator of immunoglobulin production. BLyS transgenic mice develop severe B cell hyperplasia and autoimmune lupus-like disease. Receptors for BLyS include BCMA, TACI, and BR3. BLyS and APRIL can form active heterotrimers, present in serum from patients with systemic immune-based rheumatic diseases.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of human BLyS-transfected cells. Use at 5 µL (0.25 µg) per test (final volume 100 µL). Cell number should be empirically determined (10⁵–10⁸ cells/test).

633–647 nm excitation, 660 nm emission, Red Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

BAFF is a membrane-bound cytokine expressed by B cell lineage cells. The soluble form binds to TNFRSF13B/TACI, TNFRSF17/BCMA, and TNFRSF13C/BAFFR receptors. It acts as a potent B cell activator, promoting proliferation and differentiation. BAFF is also expressed by T lymphocytes, promoting T cell activation and survival. Isoforms with alternative splicing and glycosylation variants have been identified.
