Product Specific Information

Description:
The 16-10A1 monoclonal antibody reacts with mouse CD80 (B7-1), a 55 kDa member of the Ig superfamily. CD80 is expressed by macrophages, dendritic cells, and activated B cells. Activated T cells also express this antigen. CD80 binds with high affinity to CD28 and CD152 (CTLA-4) on T cells, facilitating T-B cell communication and activation.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of stimulated mouse splenocytes. Recommended ≤1 µg per test (in 100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize concentration empirically.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be fixed using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD80 (B7-1) and CD86 (B7-2) are ligands for T cell costimulatory receptor CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on antigen-presenting cells and are essential for T cell activation. CD80 expression is induced on activated B cells, EBV-transformed B cell lines, Burkitt’s lymphoma cells, T cells, and monocytes.
CD80 provides regulatory signals for T lymphocytes via CD28 and CTLA-4 binding.
Diseases associated with CD80 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
