Target Information
The cytochrome P450 (P450) superfamily of enzymes is one of three enzyme systems that metabolize the fatty acid arachidonic acid (AA) into regulators of vascular tone. P450 enzymes are monooxygenases requiring cofactors such as NADPH and P450 reductase. Over 200 genes encode P450s.
Epoxygenases convert AA to epoxyeicosatrienoic acids (EETs), while omega-hydroxylases produce 19- and 20-hydroxyeicosatetraenoic acids (HETEs).
In addition to fatty acid metabolism, P450s metabolize many drugs and toxins. Cytochrome P450 3A4 is highly expressed in the liver and small intestine and can be induced by barbiturates, glucocorticoids, and rifampicin.
