Target Information

ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activity, playing a key role in ER-based stress signaling.

IRE1 acts as a sensor for unfolded proteins in the ER and initiates transmittance of the unfolded protein signal to the nucleus by splicing XBP1 mRNA, converting it into a potent transcriptional activator for the unfolded protein response.

Null mutants are viable, but IRE1 is essential under stress conditions that cause unfolded proteins to accumulate in the ER. Depletion of IRE1p through dominant negative expression results in apoptosis under such stress conditions.
