Target Information
Myelin Protein Zero (MPZ) is a Type 1 integral membrane glycoprotein that mediates adhesion of spiraling wraps of the myelin sheath, ensuring stable synaptic transmission. MPZ constitutes approximately 50% of total protein in the sheath scaffolding, contributing to structural integrity of peripheral myelin. It guides compact myelin wrapping through glycine zipper packing interface-dependent dimer and tetramer formation. Mutations such as G134R can disrupt multimer formation, leading to demyelinating neuropathies including Charcot-Marie-Tooth disease. The cytoplasmic domain undergoes serine and tyrosine phosphorylation, prevalent during peak nerve myelination. MPZ transcript is moderate in brain, abundant in thymus, and most abundant in white matter of the CNS.
