Target Information
Glutamate is the main excitatory neurotransmitter in the brain. It acts on ligand-gated receptor channels such as NMDA, AMPA, and kainate receptors, which mediate fast excitatory synaptic transmission. Additionally, glutamate regulates enzymes producing second messengers via G-protein-coupled receptors known as metabotropic glutamate receptors (mGluRs).
Group-I receptors, including mGluR1a and mGluR5, stimulate phospholipase C, increasing phosphoinositide turnover and calcium release from internal stores.
Group-II (mGluR2, mGluR3) and Group-III receptors are coupled to inhibition of adenylyl cyclase.
